I'm looking for best practices for backing up a ZFS based system to a cloud provider like Backblaze B2. I'm very new to ZFS and never used rclone, so I'm worry to miss essential non-trivial steps of configurations.

rclone is not a backup software. It is utility tool to access files in cloud.
It can be used as some part of DYI backup solution but if as you say you are new to these things I suggest you look at something more suitable.
For free cloud backup software I would suggest restic/rustic or kopia.
